dedicator of cytokinesis protein 1; 180 kD protein downstream of CRK; DOCK180 (DOCK1)
Function:
1) cytoskeletal rearrangements for phagocytosis of apoptotic cells & cell motility
2) guanine nucleotide exchange factor (GEF)
a) activates Rac Rho small GTPases by exchanging bound GDP for free GTP
b) GEF activity may be enhanced by ELMO1
3) interacts with the SH3 domains of CRK & NCK2
4) interacts with nucleotide-free RAC1 via its DHR-2 domain
5) interacts with ELMO1, ELMO2, ELMO3 via its SH3 domain
6) interacts with RAC1 & BAI1
Structure:
- the DHR-2 domain is necessary & sufficient for GEF activity
- belongs to the DOCK family
- contains 1 DHR-1 domain
- contains 1 DHR-2 domain
- contains 1 SH3 domain
Compartment:
- cytoplasm
- recruited to membranes via its interaction with PIP3 (phosphatidylinositol 3,4,5-triphosphate)
Expression:
- expressed in placenta, lung, kidney, pancreas, ovary > thymus, testes, colon
General
