Function: - cell surface receptor - modulates signaling cascades - mediates tyrosine phosphorylation of target MAP kinases - interacts with PTPN6 & PTPN11 Structure: - highly N-glycosylated - glycosylation varies between cell types - contains 1 ITIM motif - phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases - contains 1 C-type lectin domain Compartment: - cell membrane - ligand binding leads to internalization Alternative splicing: named isoforms=5 Expression: - detected in normal myeloid cells & in acute myeloid leukemia cells - detected in neutrophils, eosinophils, monocytes & dendritic cells - detected in spleen macrophage-rich red pulp & in lymph node (at protein level) - detected in peripheral blood leukocytes, dendritic cells, bone marrow, monocytes, mononuclear leukocytes & macrophages Pathology: - down-regulated in activated leukocytes recruited to a site of inflammation
