Myb proto-oncogene protein (C-myb, MYB)
Function:
- transcriptional activator
- DNA-binding protein that specifically recognizes sequence 5'-YAAC[GT]G-3'
- role in control of proliferation & differentiation of hematopoietic progenitor cells
- binds MYBBP1A
- interacts with HIPK2 & NLK
- ubiquitinated; mediated by SIAH1 & leading to its subsequent proteasomal degradation (probable)
- phosphorylated by NLK on multiple sites, which induces proteasomal degradation (putative)
Structure:
- comprised of 3 domains
a) N-terminal DNA-binding domain
b) centrally located transcriptional activation domain
c) C-terminal domain involved in transcriptional repression
- contains 3 HTH myb-type DNA-binding domains
Compartment: nucleus
Alternative splicing: named isoforms=6
Expression:
- normally expressed in immature hematopoietic cell lineages
Note: short 1/2 life
