Coxsackievirus & adenovirus receptor; CAR; hCAR; CVB3-binding protein; Coxsackievirus B-adenovirus receptor; HCVADR (CXADR CAR)

Function: - component of the epithelial apical junction complex - homophilic cell adhesion molecule (putative) - recruits MPDZ to intercellular contact sites - probably involved in transepithelial migration of neutrophils through adhesive interactions with AMICA1/JAML located in the plasma membrane of neutrophils - palmitoylated on Cys-259 &/or Cys-260; required for proper localization to the plasma membrane - monomer; probably homodimer formed by 2 molecules on adjacent cells - interacts with LNX, MPDZ, BAIAP1, DLG4, PRKCABP, TJP1 & CTNNB1 - secreted isoform 3, isoform 4 & isoform 5 can interact with the extracellular domain of the receptor - interacts with DNAJC7 - probably part of a complex containing CXADR, HSP90AA1 & DNAJC7 (putative) Structure: - N-glycosylated (putative) - the Ig-like C2-type 1 domain probably mediates homodimerization & interaction with JAML - the PDZ-binding motif mediates interaction with MPDZ & BAIAP1 - contains 2 Ig-like C2-type domains Compartment: - isoforms 1 & 2: - cell junction, tight junction, adherens junction - basolateral cell membrane - single-pass type 1 membrane protein - in epithelial cells localizes to apical junction complex composed of tight junction & adherens junction - in airway epithelial cells localized to basolateral membrane but not to apical surface - isoforms 3,4,5: secreted Alternative splicing: named isoforms=5 Expression: - expressed in pancreas, brain, heart, small intestine, testis, prostate - lower expession in liver & lung - isoform 5 is ubiquitously expressed - isoform 3 is expressed in heart, lung & pancreas - in skeletal muscle, isoform 1 is found at the neuromuscular junction & isoform 2 is found in blood vessels - in cardiac muscle, isoform 1 & isoform 2 are found at intercalated disks - in heart a) expressed in subendothelial layers of the vessel wall b) not found in the luminal endothelial surface c) expression is elevated in hearts with dilated cardiomyopathy Pathology: - interacts with adenovirus subgroup A, C, D, E & F fiber proteins as well as coxsackievirus B1, B2, B3, B4, B5 & B6 capsid proteins & acts as a receptor for these viruses
