Function: - organization of cell structure - associated with actin/integrin focal contacts - may associate with Shank at the post synaptic density [3] - substrate for the pp60v-src tyrosine kinase - role in synaptic plasticity by mediating the local assembly of cytoskeleton & interacting with the NMDA receptor & PSD-95 [4] - SH3 motif may function as a binding region to cytoskeleton - interacts with SHANK2 & SHANK3 via its SH2 domain - interacts with FGD1 (putative) - interacts with PLXDC2 Structure: - contains 7 cortactin repeats - contains 1 SH3 domain Compartment: - cytoplasm, cytoskeleton (putative) - cell projection, lamellipodium (putative) - cell projection, ruffle (putative) - associated with membrane ruffles & lamellipodia (putative) Pathology: - Tyr phosphorylation in transformed cells may contribute to cellular growth regulation & transformation
