Function: - chloride transport protein - initially identified as voltage-gated chloride channel - the presence of the conserved gating Glu suggests that it functions as antiporter Structure: - N-glycosylated on several asparagine residues - belongs to the chloride channel (TC 2.A.49) family - contains 2 CBS domains Compartment: - endosome membrane; multi-pass membrane protein - detected in detergent-resistant lipid rafts Alternative splicing: named isoforms=5; A-E Expression: - testis, ovary, small intestine, brain & skeletal muscle - low level expression in aortic & coronary vascular smooth muscle cells, & aortic endothelial cells - isoform C is only detected in kidney Notes: - the CLC channel family contains both chloride channels & proton-coupled anion transporters that exchange chloride or another anion for protons - the presence of conserved gating Glu is typical for family members that function as antiporters (putative)
