cAMP responsive element-binding protein 3-like protein 2 (BBF2 human homolog on chromosome 7, CREB3L2, BBF2H7)
Function:
1) transcriptional activator
2) may act during endoplasmic reticulum stress late phase by activating unfolded protein response target genes
3) may play a role in preventing accumulation of unfolded proteins in damaged neurons
4) in vitro, binds to the cAMP response element (CRE) & activates transcription through CRE
5) binds DNA as a dimer
6) controlled by regulated intramembrane proteolysis
7) following ER stress a fragment containing the cytoplasmic transcription factor domain is released by proteolysis
Structure:
- belongs to the bZIP family, ATF subfamily.
- contains 1 bZIP domain
Compartment:
- endoplasmic reticulum membrane
- processed form: nucleus
- under ER stress the cleaved N-terminal cytoplasmic domain translocates into the nucleus
Alternative splicing: named isoforms=3
Expression:
- widely expressed
- highest levels in placenta, lung, spleen & intestine
- lowest levels in heart, brain, skeletal muscle, thymus, colon & leukocytes tissues, weakest expression is detected in brain & heart
Pathology:
- chromosomal translocation t(7;16)(q33;p11) involving CREB3L2 with FUS is found in low grade fibromyxoid sarcoma
