Function: - interacts directly with CLOCK to form BMAL2-CLOCK transactivator - BMAL2-CLOCK heterodimers activate E-box element (3'- CACGTG-5') transcription - in umbilical vein endothelial cells, activates SERPINE1 through E-box sites, this transactivation is inhibited by PER2 & CRY1. - component of the circadian core oscillator - iinteracts with NPAS2, HIF1A Structure: - contains 1 basic helix-loop-helix domain (bHLH) - contains 1 PAC domain (PAS-associated C-terminal) - contains 2 PAS domains (PER-ARNT-SIM) Commpartment: nucleus Alternative splicing: named isoforms=8 Expression: - expressed in fetal brain - expressed in brain & placenta - lower levels in heart, liver, thymus, kidney, lung - expressed in endothelial cells & neuronal cells of the suprachiasmatic nucleus - detected in endothelial cells of the heart, lung & kidney - in the brain, specifically expressed in the thalamus, hippocampus & amygdala - constitutively expressed - no circadian rhythm expression pattern
