centromere/kinetochore protein zw10 homolog (ZW10)

Function: - essential component of the mitotic checkpoint, which prevents cells from prematurely exiting mitosis - required for assembly of the dynein-dynactin & MAD1-MAD2 complexes onto kinetochores - involved in regulation of membrane traffic between the Golgi & the endoplasmic reticulum - associated with a SNARE complex through direct interaction with RINT1/TIP20L bound to BNIP1/SEC20L - component of the RZZ complex - interacts with C19orf25, KNTC1, & ZWINT Structure: belongs to the ZW10 family Compartment: - cytoplasm - endoplasmic reticulum membrane - peripheral membrane protein - chromosome, centromere, kinetochore - cytoskeleton, spindle - dynamic pattern of localization during the cell cycle - in most cells at interphase, present diffusely in cytoplasm - in prometaphase, associated with the kinetochore - at metaphase, detected both at the kinetochores &, most prominently, at the spindle, particularly at the spindle poles - in very early anaphase, detected on segregating kinetochores - in late anaphase & telophase, accumulates at the spindle midzone Expression: - widely expressed - no significant variation in expression during cell cycle - overexpression as well as silencing of ZW10 disrupts morphology of the ER-Golgi intermediate compartment & slows ER-Golgi transport
