Function: - E3 ubiquitin-protein ligase - mediates ubiquitination & subsequent proteasomal degradation of myosin regulatory light chain (MRLC), LDLR, VLDLR & LRP8 - proteasomal degradation of MRLC inhibits neurite outgrowth in presence of NGF by counteracting the stabilization of MRLC by saposin-like protein (CNPY2/MSAP) & reducing CNPY2-stimulated neurite outgrowth - acts as a sterol-dependent inhibitor of cellular cholesterol uptake by mediating ubiquitination & subsequent degradation of LDLR - protein modification; protein ubiquitination - autoubiquitinated - interacts with myosin regulatory light chain (MRLC) & TMEM4 Structure: - the RING domain mediates ubiquitination & the neurite outgrowth inhibitory activity - contains 1 FERM domain - contains 1 RING-type Zn+2 finger Compartment: cytoplasm (probable) Alternative splicing: named isoforms=2 Expression: - ubiquitously expressed - expressed in fetal tissues - higher expression in placenta & fetal lung - induced expression is directly activated by NR1H2 & NR1H3 - expression is indirectly induced by LDL - not sterol-response element-binding protein (SREBP) dependent
