Function: - transcriptional repressor - probably represses transcription by binding directly the 5'-CTGTCAA-3' DNA sequence or by interacting with TGF-beta activated SMAD proteins - probably represses transcription via the recruitment of histone deacetylase proteins - the C-terminal part is phosphorylated in response to EGF signaling by the Ras/MAPK pathway - interacts with the transcriptional modulator SMAD3 & the histone deacetylase HDAC1 Structure: - belongs to the TALE/TGIF homeobox family - contains 1 homeobox DNA-binding domain Compartment: - nucleus - excluded from nucleoli Expression: - widely expressed - highly expressed in heart, kidney & testis - weakly expressed in brain & prostate Pahtology: - TGIF2 is amplified & overexpressed in several ovarian cancer cell lines
