Function:
1) involved in mitotic spindle checkpoint/mitotic regulation
2) binds the telomeric double-stranded TTAGGG repeat & negatively regulates telomere length
3) important for maintaining mitotic checkpoint in response to DNA damage
4) interacts with ATM, TINF2, TNKS1, TNKS2, PINX1, NEK2, MAPRE1
5) phosphorylated preferentially on Ser-219 by ATM kinase in response to ionizing DNA damage.
6) ADP-ribosylation by TNKS1 or TNKS2 diminishes its ability to bind to telomeric DNA
7) found in a complex with POT1, TINF2 & TNKS1.
Structure:
- homodimer
- the acidic N-terminal domain binds to the ankyrin repeats of TNKS1 & TNKS2
- C-terminal domain binds microtubules
Compartment:
- nuclear
- colocalizes with telomeric DNA in interphase & metaphase cells
- located at chromosome ends during metaphase
- associates with the mitotic spindle
Expression:
- highly expressed & ubiquitous
- isoform Pin2 predominates
- Pin2 expression is tighly regulated during the cell cycle
- levels of Pin2 are low in G1 & S phase & increase during G2 phase & mitosis
Notes:
- Pin2 is identical to TRF1 except for a 20 amino acid deletion
