Indications: - treatment of childhood solid tumors - androgen-independent metastatic prostate cancer Dosage: Tabs: 0.25 mg, 1 mg Mechanism of action: - inhibits poly(ADP-ribose) polymerase (PARP) - prevents PARP-mediated DNA repair of single-strand DNA breaks via the base-excision repair pathway - enhances the accumulation of DNA strand breaks, promotes genomic instability & eventually leads to apoptosis - PARP catalyzes post-translational ADP-ribosylation of nuclear proteins that signal & recruit other proteins to repair damaged DNA & is activated by single-strand DNA breaks
