Function:
- mucin like protein
- may play role in tumor progression
- cell migration &/or actin cytoskeleton organization
- lung cell proliferation & alveolus formation at birth
- induces platelet aggregation
- when expressed in keratinocytes, induces changes in cell morphology with transfected cells showing an elongated shape, numerous membrane protrusions, major reorganization of the actin cytoskeleton, increased motility & decreased cell adhesion
- does not have any effect on folic acid or amino acid transport
- does not function as a water channel or as a regulator of aquaporin-type water channels
Structure:
- belongs to the podoplanin family
- O-glycosylation (sialic acid) necessary for platelet aggregation
- the N-terminus is blocked (putative)
Compartment:
- localized to actin-rich microvilli & plasma membrane projections (filopodia, lamellipodia, ruffles)
- apical plasma membrane of endothelium
Alternative splicing: named isoforms=2
Expression:
- expressed in placenta, lung, skeletal muscle, brain > brain, kidney, liver
- expressed in:
- apical plasma membrane of endothelium
- alveolar epithelium
Pathology:
- up-regulated in colorectal tumors
- expressed in 25% of early oral squamous cell carcinomas
