Function: - histone methyltransferase - specifically trimethylates Lys-20 of histone H4 - histone H4 Lys-20 trimethylation represents a specific tag for epigenetic transcriptional repression - mainly functions in pericentric heterochromatin regions, & plays role in establishment of constitutive heterochromatin in these regions - SUV420H1 is targeted to histone H3 via its interaction with RB1 family proteins (RB1, RBL1 & RBL2) - interacts with HP1 proteins CBX1, CBX3 & CBX5 S-adenosyl-L-methionine + histone L-lysine S-adenosyl-L-homocysteine + histone N(6)-methyl-L-lysine Compartment: - nucleus - associated with pericentric heterochromatin - CBX1 & CBX5 are required for localization to pericentric heterochromatin Alternative splicing: named isoforms=3 Structure: - belongs to the histone-lysine methyltransferase family, Suvar4-20 subfamily - contains 1 SET domain
