Function: 1) regulates actin filament dynamics 2) dephosphorylates & activates cofilin (disassembles actin) 3) LIMK1 phosphorylates & deactivates cofilin 4) interacts with 14-3-3 proteins YWHAB, YWHAG, YWHAQ, & YWHAZ 5) inhibits phosphatase activity 6) inhibitory phosphorylation by PAK4 promotes binding to YWHAZ 7) dephosphorylated & activated by calcineurin A Structure: - belongs to the protein-tyrosine phosphatase family - contains 1 tyrosine-protein phosphatase domain Compartment: - cytoplasm - also recruited to actin rich-membrane protrusions, lamellipodia for cell locomotion - also localized to the cleavage furrow & the midbody during cytokinesis Alternative splicing: - named isoforms=5 - at least one isoform may be produced at very low levels due to a premature stop codon in the mRNA, leading to nonsense-mediated mRNA decay
