lactate dehydrogenase (LDH) in serum

Reference values: - 105-333 IU/L [8] - Kodak Ektachem - Male: 259- 613 U/L - Female: 259- 613 U/L Clinical significance: Lactate dehydrogenase is an enzyme present in the cytosol of the human cells; it catalyzes the reversible reduction of pyruvate to lactate using NADH. In myocardial infarction cases, LD begins to rise within about 12 hours after infarction, & usually returns to normal after 2 to 5 days. Increases: - neoplasms - hypoxia associated with cardiopulmonary diseases - myocardial infarctions - hemolytic anemias - megaloblastic anemias - hepatic cirrhosis - renal infarction - trauma - muscle damage - muscular dystrophy - shock - hypotension Decreases: -> pharmaceutical agents: a) in vivo effects -> clofibrate b) chemical interferences - oxalate, theophylline Principle: - see lactate dehydrogenase in body fluid Specimen: Sample preparation: Collect the specimen by standard venipuncture technique. Heparin may be used as an anticoagulant for plasma specimens. Remove serum or plasma promptly from the clot or cells and analyze as soon as possible. EDTA, potassium oxalate & sodium fluoride should not be used as anticoagulants. Serum or heparin specimens produce similar LDH results on the Kodak Ektachem Analyzer. Some other methods, however, have shown substantial differences between serum or plasma results due to contamination by platelets in plasma separated by low speed centrifugation. The Kodak Ektachem Clinical Chemistry Slides (LDH) are insensitive to LDH contained within intact platelets, therefore, LDH results in comparative methods may not agree with the Kodak Ektachem Analyzer for heparin plasma specimens. Sample requirements: Minimum- 0.5 mL serum or plasma. Optimum: 1.0 mL serum or plasma.
