Function: - filament-forming cytoskeletal GTPase (putative) - may play a role in cytokinesis (putative) - may play a role in platelet secretion - isoform 6, but not the other isoforms, is required for induction of cell death mediated by TGF-beta & by other apoptotic stimuli - septins polymerize into heterooligomeric protein complexes that form filaments, & can associate with cellular membranes, actin filaments & microtubules - GTPase activity is required for filament formation (putative) - interacts with SEPT8 - in a mesenchymal cell line, interacts with SEPT9 isoform 2 Structure: belongs to the septin family Compartment: - cytoplasm (putative) - cytoskeleton (putative) - in platelets, found in areas surrounding alpha-granules - isoform 6: - mitochondrion, nucleus - predominantly localized in the mitochondria under resting conditions - translocates into the nucleus after TGF-beta treatment & induction of apoptosis Alternative splicing: named isoforms=6 Expression: - widely expressed in adult & fetal tissues - highest expression in adult brain (at protein level), heart, liver & adrenal gland & fetal heart, kidney, liver & lung - also expressed in colorectal cancers & malignant melanomas - expressed in platelets
