radical S-adenosyl methionine domain-containing protein 2 (virus inhibitory protein, endoplasmic reticulum-associated, interferon-inducible, viperin, cytomegalovirus-induced gene 5 protein, RSAD2, CIG5)
Function:
- role in antiviral defense
- inhibits infection by human cytomegalovirus, down-regulating expression of a number of viral proteins including glycoprotein B, pp28 & pp65
- inhibits replication of hepatitis C virus
Cofactor:
- binds 1 4Fe-4S cluster
- cluster is coordinated with 3 cysteines & an exchangeable S-adenosyl-L-methionine
Structure: belongs to the RSAD2 family
Compartment:
- endoplasmic reticulum, Golgi apparatus
- probably associates with the cytosolic side of the endoplasmic reticulum
- infection with human cytomegalovirus (HCMV) causes relocation to the Golgi & to cytoplasmic vacuoles which also contain HCMV proteins glycoprotein B & pp28
Expression:
- induced by LPS, interferon alpha &, to a lesser extent, interferon gamma
- little or no induction by interferon gamma is observed in monocytic cell lines
- induced by infection with human cytomegalovirus (HMCV), hepatitis C virus, yellow fever virus & Sendai virus
- induction by HMCV is mediated by glycoprotein B
Pathology:
- expressed in the endothelium of atherosclerotic arteries but not in normal arteries (at protein level)
