- implicated in the assembly of focal adhesions & actin stress fibers
- regulates a signal transduction pathway linking plasma membrane receptors to the assembly of focal adhesions & actin stress fibers
- may be an activator of PLCE1
- activated by ARHGEF2, which promotes the exchange of GDP for GTP
- interacts with RGNEF (putative)
- binds PRKCL1, ROCK1 & ROCK2
- interacts with ARHGEF2, ARHGEF3, NET1 & RTKN
- interacts with PLCE1 & AKAP13

Structure:
- basic-rich region is essential for yopT recognition & cleavage
- belongs to the small GTPase superfamily, Rho family

Pathology:
- serves as a target for the yopT cysteine peptidase from Yersinia pestis & Yersinia pseudotuberculosis
- cleaved by yopT protease when the cell is infected by Yersinia; this removes the lipid attachment, & leads to its displacement from plasma membrane & to subsequent cytoskeleton cleavage
- substrate for botulinum ADP-ribosyltransferase
