restriction fragment length polymorphism (RFLP)
Clinical significance:
- polymorphisms may arise from single base mutations, deletions or insertion of DNA
- RFLP analysis is useful for genetic linkage studies to establish carrier (heterozygous) state in kindred of individuals known to be affected with specific genetic disorders
Procedure:
- analysis of DNA polymorphisms by agarose gel electrophoresis after cleavage of DNA with restriction endonucleases
