REST corepressor 1 (protein CoREST, RCOR1, KIAA0071, RCOR)
Function:
1) component of the BHC complex
2) in the BHC complex, RCOR1 recruits MeCP2 & SUV39H1
3) role in demethylation of Lys-4 of histone H3 by promoting demethylase activity of AOF2/LSD1 on core histones & nucleosomal substrates
4) protects AOF2/LSD1 from the proteasome
5) SANT domains may bridge the nucleosomal substrates & the demethylase AOF2/LSD1
Structure:
1) belongs to the CoREST family
2) contains 1 ELM2 domain
3) contains 2 SANT domains
Pathology:
- interacts directly with herpes virus HSV-1 ICP0 protein, leading to the disruption of the BHC complex, thereby preventing the BHC complex from repressing transcription of viral genes
- upon infection by HSV-1, partially translocated into cytoplasm
- phosphorylated by HSV-1 protein kinases
