pumilio homolog 2 (pumilio-2, PUM2, KIAA0235, PUMH2)

Function: 1) sequence-specific RNA-binding protein 2) regulates translation & mRNA stability by binding the 3' untranslated regions (UTRs) of mRNA targets 3) role in proliferation & self-renewal of stem cells by regulating translation of key transcripts (putative) 4) interacts with DAZ, DAZL & NANOS1 via its pumilio repeats 5) binds to RNA consensus sequence, related to Nanos Response Element (NRE), a 16 bp sequence found in the 3'UTR of the Drosophila hb mRNA 6) binds to the NRE Structure: - homodimer - contains 8 pumilio repeats - the pumilio repeats mediate the association with RNA by packing together to form a right-handed superhelix that approximates a half donut - the number as well as the specific sequence of the pumilio repeats determine the specificity for target mRNAs - contains 1 PUM-HD domain Compartment: cytoplasm Alternative splicing: named isoforms=3 Expression: - expressed in testis, ovary, stem cells, germ cells > brain, heart, kidney, liver, muscle, placenta, intestine, stomach - expressed in cerebellum, corpus callosum, caudate nucleus, hippocampus, medulla oblongata, putamen. - expressed in all fetal tissues - dwn-regulated in keratinocytes upon UV B radiation
