protein N-terminal asparagine amidohydrolase; protein NH2-terminal asparagine amidohydrolase; PNAA; protein NH2-terminal asparagine deamidase; PNAD; protein N-terminal Asn amidase; protein NTN-amidase (NTAN1)
Function:
1) side-chain deamidation of N-terminal Asn to Asp
2) ubiquitin-dependent turnover of intracellular proteins that initiate with Met-Asn
a) these proteins are acetylated on the retained initiator Met & can subsequently be modified by the removal of N-acetyl methionine by acylaminoacid hydrolase (AAH)
b) conversion of the resulting N-terminal Asn to Asp by PNAD renders the protein susceptible to arginylation, polyubiquitination & degradation as specified by the N-end rule
3) does NOT act on substrates with internal or C-terminal Asn
4) does NOT act on Gln residues in any position
