FYVE finger-containing phosphoinositide kinase; 1-phosphatidylinositol-4-phosphate 5-kinase; PtdIns(4)P-5-kinase p235 (PIP5K, PIKFYVE)

Function: - supports the intracellular PIP pool & to a lesser extent, the PtdIns(4,5)P2 pool - generates PIP from PI &, to a lesser extent, PtdIns(4,5)P2 pool from PtdIns4P - indications that it phosphorylates the D-5 rather than the D-4 position - role in endosome-related membrane trafficking (putative) ATP + 1-phosphatidyl-1D-myo-inositol 4-phosphate ADP + 1-phosphatidyl-1D-myo-inositol 4,5-bisphosphate Structure: - contains 1 DEP domain - contains 1 FYVE-type Zn+2 finger - contains 1 PI5K domain Compartment: - endosome membrane - mainly associated with membranes of the late endocytic pathway Pathology: - defects in PIP5K3 are the cause of corneal fleck dystrophy
