pim1 proto-oncogene protein

Function: - Pim-1 interacts with & may play a role in promoting complex formation between NuMA, HP1-beta, dynein & dynactin which is necessary for mitosis - during prometaphase, pim-1 interacts with NuMA & centromere associated protein HP1 - Pim-1 phosphorylates NuMA [4]. - Hsp90-alpha & hsp90-beta interact with pim-1 & affect its stablization & function. - Pim-1 phosphorylation of cdc25A upregulates phosphatase activity - Pim-1 is regulated by various cytokines including GM-CSF, G-CSF, interleukin-3, interferon-alpha & interleukin-6 Compartment: - during the cell cycle, pim-1 moves from the nucleus & cytoplasm in interphase to the spindle poles during mitosis - in late metaphase/early anaphase pim-1 associates with the spindle & moves to the cleavage furrow during telophase Pathology: - overexpressed in a subset of hematolymphoid malignancies - also expressed in high levels in malignant prostate tissue, but absent in metastatic tissue Note: - there is a pim1 gene in Saccharomyces pombe that is a homolog of RCC1 - the product of the yeast gene is a serine/threonine kinase implicated in coupling S -> M phase transition in the cell cycle
