Function: 1) adapter protein located at the actin cytoskeleton 2) promotes cell attachment 3) role in migratory capacity of epithelial cells 4) interacts with alpha-actinins ACTN1 & ACTN4, FLNA & MYH9 Structure: - contains 1 LIM zinc-binding domain - contains 1 PDZ domain (DHR domain) Compartment: - isoforms 1 & 2: cytoplasm, cytoskeleton - isoform 3: nucleus. may be partially nuclear - isoform 1 colocalizes with beta-1 integrin (ITGB1) & alpha-actinin but not with paxillin Alternative splicing: named isoforms=4 Pathology: - overexpression enhances cell adhesion to collagen & fibronectin & suppresses anchorage independent growth - may contribute to tumor cell migratory capacity
