Function: - growth factor - role in regulation of embryonic development, cell proliferation, cell migration, survival & chemotaxis - potent mitogen for cells of mesenchymal origin - required for - normal proliferation & recruitment of pericytes & vascular smooth muscle cells in the central nervous system, skin, lung, heart & placenta - normal blood vessel development - normal development of renal glomeruli - role in wound healing - signaling is modulated by the formation of heterodimers with PDGFA - homodimer; antiparallel disulfide-linked dimer - heterodimer with PDGFA; antiparallel disulfide-linked dimer - PDGFB homodimer interacts with PDGFRA & PDGFRB homodimers, & with heterodimers formed by PDGFRA & PDGFRB - the heterodimer composed of PDGFA & PDGFB interacts with PDGFRB homodimers, & with heterodimers formed by PDGFRA & PDGFRB - interacts with XLKD1 (putative) Structure: - belongs to the PDGF/VEGF growth factor family Compartment: - secreted - released by platelets upon wounding Expression: - expressed at high levels in the heart, brain (substantia nigra), placenta & fetal kidney - expressed at moderate levels in the brain (hippocampus), skeletal muscle, kidney & lung Pathology: - chromosomal translocation t(17;22)(q22;q13) involving PDGFB with COL1A1 is found in dermatofibrosarcoma protuberans Pharmacology: - available under the name Regranex (Ortho-McNeil) - used to promote healing in neuropathic diabetic foot ulcers
