NADPH oxidase 5 (NOX5)
Function:
1) Ca+2-dependent NADPH oxidase subunit
2) generates superoxide
3) Ca+2-dependent proton channel
- activate by Ca+2
4) may regulate redox-dependent processes in lymphocytes & spermatozoa
5) role in cell growth & apoptosis
6) down-regulated by TGFB1
Cofactor: FAD, Mg+2
Inhibition:
- inhibited by diphenylene iodonium
Kinetic parameters:
- KM=1.06 uM for Ca+2
Compartment: membrane
Alternative splicing: named isoforms=5
Expression:
- expressed in pachytene spermatocytes of testis, lymphocyte-rich areas of spleen & lymph nodes > ovary, placenta, pancreas, cardiac fibroblasts, B-cells, prostate malignant cells, isoform 3 expressed in spleen, isoform 4 expressed in testis
- expressed in fetal tissues
Pathology:
1) associated with increased proliferative capacity & diminished apoptosis
2) implicated in transformation of Barrett esophagus, melanoma, prostate cancer
