Function: 1) NOH-1S is a voltage-gated proton channel a) mediates H+ currents of resting phagocytes & other tissues b) participates in the regulation of cellular pH c) blocked by Zn+2 2) NOH-1L is a pyridine nucleotide-dependent oxidoreductase a) generates superoxide b) conduct H+ ions as part of its electron transport Cofactor: NADP, FAD Structure: - contains 1 ferric oxidoreductase domain, residues 54-545; oxidase activity is potentiated by NOXO1, via residues 397-536 Alternative splicing: named isoforms=3 Expression: - NOH-1L is expressed in colon, uterus, prostate, colon carcinoma, but NOT in peripheral blood leukocytes - NOH-1S is detected only in colon & colon carcinoma cells
