NF-kappa-B inhibitor-interacting Ras-like protein 1; I-kappa-B-interacting Ras-like protein 1; Kappa B-Ras protein 1; KappaB-Ras1 (NKIRAS1, KBRAS1)
Atypical ras-like protein.
Function:
1) regulates NF-kappa-B activity
2) inhibits degradation of NFKBIB
3) inhibits phosphorylation of NFKBIB (putative)
4) inhibits nuclear localization of RELA NF-kappa-B subunit
5) both GTP- & GDP-bound forms block phosphorylation of NFKBIB
6) cytoplasmic retention of p65/RELA NF-kappa-B subunit
7) interacts with PEST domain in both NFKBIA & NFKBIB in vitro
8) probably only interacts with NFKBIB in vivo [2]
9) in COS cells, KBRAS1 associated efficiently with IkB-beta, poorly with IkB-alpha
10) forms a complex with NFKBIB & NF-kappa-B
11) interacts with c-Rel
Structure:
- unlike other ras family proteins, conserved Gly & Gln in positions 13 & 65 are replaced by Leu in KBRAS1 & KBRAS2
- KBRAS1 & KBRAS2 resemble constitutively active, oncogenic forms of Ras
- belongs to the small GTPase superfamily, Ras family, KappaB-Ras subfamily
