Function:
1) growth suppressor, facilitates entry into cell cycle arrest
2) functionally similar to Rb protein, binds to & represses activity of cell-cycle-promoting proteins & transcription factor E2F
3) interacts with p53 & works in an additive manner to inhibit cell growth
4) functions as transcription factor & binds directly to specific guanosine-rich DNA sequences
5) binds to the transactivation domains of E2F1 & p53.
6) interacts with nucleobindin 1 & nucleobindin 2

- contains 1 MAGE domain
Compartment:
- nucleus, neural perikarya
- translocates to nucleus of postmitotic neurons & interacts with nuclear matrix
Expression:
- almost ubiquitous
- expressed in fetal brain, lung, liver & kidney, in adult heart, brain, placenta, lung, liver, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis, ovary, small intestine, colon
- not detected in peripheral blood leukocytes
- in brain, restricted to post-mitotic neurons
Pathology:
- binds to & represses activity of cell-cycle-promoting proteins SV40 large T antigen, adenovirus E1A
- defects in NDN may be a cause of the Prader-Willi syndrome
