NACHT, LRR & PYD-containing protein 2; PYRIN domain & NACHT domain-containing protein 1; PYRIN-containing APAF1-like protein 2; nucleotide-binding site protein 1 (NALP2, NLRP2, NBS1, PAN1, PYPAF2)

Function: 1) suppresses TNF- & CD40-induced NFKB1 activity via IKK complex, by inhibiting NFKBIA degradation induced by TNF 2) when associated with PYCARD, activates CASP1, leading to secretion of mature proinflammatory cytokine IL1B 3) component of the inflammasome 4) interacts with CHUK, IKBKB & IKBKG, 5) interacts with PYCARD & DAPIN domain of NAPL1, but NOT full-length protein 6) DAPIN domain is necessary & sufficient for suppression of NFKB1 activation induced by TNF & for inducing IL1B secretion in association with caspase-1 - DAPIN domain is involved in interaction with PYCARD 7) isolated NACHT domain interacts with CARD8, but interaction not observed for full-length protein (autoinhibition?) Structure: - contains 1 DAPIN domain contains 9 LRR repeats (leucine-rich repeats) contains 1 NACHT domain Compartment: cytoplasm Alternative splicing: named isoforms=4 Expression: - expressed in lung, placenta, thymus > ovary, intestine, brain - induced by interferons & lipopolysaccharide (LPS)
