MAP kinase-interacting serine/threonine kinase 1 (MKNK1, MAP kinase signal-integrating kinase 1)

Function: - may play a role in the response to environmental stress & cytokines - appears to regulate transcription by phosphorylating EIF4E, thus increasing its affinity for the 7-methylguanosine- containing mRNA cap - interacts with the C-terminal regions of EIF4G1 & EIF4G2 - also binds to dephosphorylated ERK1 & ERK2, & to the p38 kinases - phosphorylated & activated by the p38 kinases & kinases in the Erk pathway - dual phosphorylation of Thr-250 & Thr-255 activates the kinase - phosphorylation of Thr-385 activates the kinase ATP + a protein ADP + a phosphoprotein Cofactor: Mg+2 Structure: - belongs to the protein kinase superfamily, CAMK Ser/Thr protein kinase family - contains 1 protein kinase domain Compartment: - isoform 2: cytoplasm - isoform 3: cytoplasm, nucleus Alternative splicing: named isoforms=3 Expression: ubiquitous
