DNA replication licensing factor MCM6; p105MCM (MCM6)
Function:
- may be involved in control of a single round of DNA replication during S phase
- binds to chromatin during G1 phase & detaches from it during S phase as if it licenses the chromatin to replicate
- phosphorylated upon DNA damage, probably by ATM or ATR
- may interact with MCM10
- interacts with TIPIN
- forms a stable trimeric complex with CDC21 & p85MCM.
- protein BM28/Mcm2 is more loosely associated with the trimeric complex
Structure:
- belongs to the MCM family
- contains 1 MCM domain
Compartment:
- nucleus (putative)
Polymorphism:
- intronic variations in MCM6 upstream from the LCT gene are associated with adult-type hypolactasia leading to lactose intolerance, or with lactase persistence
- a non-coding variation in MCM6 affects the transcriptional regulation of the LCT gene resulting in down-regulation of lactase activity
