methyl-CpG-binding domain protein 1 (methyl-CpG-binding protein MBD1, protein containing methyl-CpG-binding domain 1, MBD1, PCM1)
Function:
1) binds CpG islands in promoters where DNA is methylated at position 5 of cytosine within CpG dinucleotides
2) binding abolished by the presence of 7-mG produced by DNA damage by methylmethanesulfonate
3) transcriptional repressor, gene silencing
4) isoforms 1 & 2 can repress transcription from unmethylated promoters - binding via third CXXC-type zinc finger
5) binds OASL, ATF7IP, CHAF1A, SUV39H1-CBX5 complex binds MGP via TRD domain
6) component of MeCP1 complex (putative)
7) up-regulated by interferon
Compartment:
- nucleus, nuclear speckle
- associated with euchromatic regions of chromosomes, pericentromeric regions on chromosome 1 & telomeric regions of several chromosomes
Alternative splicing: named isoforms=7
Expression: widely expressed
