mitogen-activated protein kinase 3 (ERK1, ERT2, p44mapk, p44erk1, insulin-stimulated MAP2 kinase, myelin basic protein [MBP] kinase E4, RSK kinase-2, MAPK3, PRKM3)
Function:
- role in both the initiation & regulation of meiosis, mitosis, & postmitotic functions in differentiated cells
- activated in by Tyr phosphorylation response to insulin & NGF
- dually phosphorylated on Thr-202 & Tyr-204, which activates the enzyme
- phosphorylates MAP2, MBP, ELK-1 EIF4EBP1
- required for initiation of translation
- phosphorylates phosphorylates SPZ1 (putative)
- phosphorylates heat-shock-factor protein 4 (HSF4)
- interacts with MORG1 (putative)
- binds to HIV-1 Nef; interaction inhibits its kinase activity
- interacts with HSF4
Structure:
- 33% homologous with p34cdc2
- TXY motif contains the Thr & Tyr whose phosphorylation activates the MAP kinases
- belongs to the protein kinase superfamily, CMGC Ser/Thr protein kinase family, MAP kinase subfamily
- contains 1 protein kinase domain
Expression:
- predominates in intestine & lung
- expression in hippocampus limited to dentate granule cells [2]
