Function: - MAP2K3 & MAP2K6 activate p38MAPKs - catalyzes concomitant phosphorylation of a Thr & a Tyr in MAP kinase p38 exclusively - interacts with Yersinia yopJ - probably activated by dual phosphorylation on Ser-207 & Thr-211 - weakly autophosphorylated - acetylation of Ser-207 & Thr-211 by Yersinia yopJ prevents phosphorylation & activation, thus blocking the MAPK signaling pathway ATP + a protein = ADP + a phosphoprotein Structure: - belongs to the protein kinase superfamily, STE Ser/Thr protein kinase family, MAP kinase kinase subfamily - contains 1 protein kinase domain Alternative splicing: named isoforms=2 Expression: - isoform 2 is only expressed in skeletal muscle - isoform 1 is expressed in skeletal muscle, heart > liver, pancreas - strongly activated by UV, anisomycin, & osmotic shock but not by phorbol esters, NGF or EGF
