mitogen activated protein kinase kinase 5 (MAPK/ERK kinase 5, MEK5, MAP2K5, MKK5, PRKMK5)

Function: - may participate together with ERK5 in a growth factor stimulated signaling cascade - specifically activates ERK5, but not other MAP kinases such as ERK2 or p38 - not phosphorylated by RAFA, RAFB or RAFC - may interact with GTPases such as CDC42 - interacts with PARD6A & MAP3K3 - forms a complex with SQSTM1 & PRKCZ or PRKCI (putative) - interacts with Yersinia yopJ - activated by phosphorylation on Ser/Thr by MEK kinases (putative) - Yersinia yopJ may acetylate Ser/Thr residues, preventing phosphorylation & activation, thus blocking the MAPK signaling pathway ATP + a protein = ADP + a phosphoprotein Structure: - OPR domain mediates interactions with SQSTM1, PARD6A & MAP3K3 (putative) - belongs to the protein kinase superfamily, STE Ser/Thr - protein kinase family, MAP kinase kinase subfamily - contains 1 OPR domain - contains 1 protein kinase domain Alternative splicing: named isoforms=3 Expression: - expressed in many adult tissues - abundant in heart & skeletal muscle
