Clinical significance: - protein bound amylase - due to its larger size, macroamylase may not be cleared as fast as the smaller non-protein bound amylase, thus levels may be elevated in the absence of pathology Increases: - macroamylasemia Methods: 1) direct identification in serum requires ultracentrifugation or chromatography 2) indirect methods: a) selective precipitation of macroamylase in a polyethylene glycol solution b) temperature sensitive amylase assay (dissociation of amylase from complexed proteins)
