Function: - binds LDL, the major cholesterol-carrying lipoprotein of plasma, & transports it into cells by endocytosis - in order to be internalized, the receptor-ligand complexes must first cluster into clathrin-coated pits - the LDL receptor binds 2 ligands, apo B-100 & apo-E, apolipoproteins with completely different sequences - interacts with LDLRAP1 - interacts with SNX17 Structure: - N- & O-glycosylated - belongs to the LDLR family - contains 3 EGF-like domains - contains 7 LDL-receptor class A domains - contains 6 LDL-receptor class B repeats - Ser/Asp/Glu triplet at C-terminal end of L-module is important for ligand-binding Compartment: membrane, clathrin-coated pit Expression: - mRNA for the LDL receptor is expressed in a variety of tissues including liver & adrenals Pathology: - defects in LDLR are the cause of familial hypercholesterolemia) - interacts with HIV-1 Tat in case of HIV-1 infection, functions as a receptor for extracellular Tat in neurons, mediating its internalization in uninfected cells - interacts with HCV E1/E2 heterodimer Note: - also see apo B-48 receptor
