HIV1 RNA viral load

Indications: - assessment of HIV1 infection - AIDS-defining condition - Kaposi sarcoma - CNS lymphoma - pneumocystis pneumonia - progressive multifocal leukoencephalopathy - screening for HIV1* * follow-up of negative HIV1 serology with HIV1 p24 antigen & HIV1 RNA improves detaction rates in high-risk populations [2] Procedure: 1) branched DNA (bDNA) assay - generally give 1/2 the HIV RNA levels of RT-PCR 2) Reverse-transcriptase polymerase chain reaction (RT-PCR) a) Roche Amplicor ultrasensitive test b) Roche TaqMan HIV-1 3) lower limit of detection 20-50 copies/mL Specimen: 1) whole blood (ACD or EDTA) 2) store at 4 degrees C Notes: - PCR primers are directed towards env, gag & pol regions of the HIV genome - it is possible to monitor infection during the latent period
