histone deacetylase 5 (HD5, antigen NY-CO-9, HDAC5 KIAA0600)
Function:
- see histone deacetylase
- deacetylation of Lys on N-terminal part of core histones (H2A, H2B, H3 & H4)
- involved in muscle maturation by repressing transcription of myocyte enhancer MEF2C
- during muscle differentiation, shuttles into the cytoplasm, allowing expression of myocyte enhancer factors
- interacts with BCOR, HDAC7, HDAC9, CTBP1, MEF2C, NCOR2, NRIP1, PHB2 & a 14-3-3 chaperone protein
- interacts with JARID1B
- phosphorylated by CaMK at Ser-259 & Ser-498
- phosphorylation is required for the export to cytoplasm
- ubiquitinated
- polyubiquitination does NOT lead to its degradation
Structure:
- nuclear export sequence mediates shuttling between nucleus & the cytoplasm (putative)
- belongs to the histone deacetylase family, type 2 subfamily
Compartment:
- nucleus, cytoplasm
- shuttles between the nucleus & the cytoplasm
- in muscle cells, shuttles into cytoplasm during myocyte differentiation
- export to cytoplasm depends on interaction with a 14-3-3 chaperone protein & is its phosphorylation at Ser-259 & Ser-498 by CaMK
Alternative splicing: named isoforms=2
Expression: ubiquitous
