histone deacetylase 2 (HDAC2)
Function:
- see histone deacetylase
- deacetylation of Lys on N-terminal part of core histones (H2A, H2B, H3 & H4)
- forms transcriptional repressor complexes by associating with MAD, SIN3, YY1 & N-COR
- interacts in the late S-phase of DNA-replication with DNMT1 in the other transcriptional repressor complex composed of DNMT1, DMAP1, PCNA, CAF1
- interacts with the non-histone region of H2AFY
- part of the core histone deacetylase complex (HDAC complex)
- component of BHC histone deacetylase complex
- component of a mSin3A corepressor complex
- part of a complex containing the core histones H2A, H2B, H3 & H4, DEK & unphosphorylated DAXX
- part of a complex containing ATR & CHD4
- forms a heterologous complex at least with YY1
- interacts with ATR, DNMT1, MINT, HDAC7, HDAC10, HCFC1, NRIP1, MJD2A/JHDM3A, SAP30, SETDB1 & SUV39H1
- interacts with the non-histone region of H2AFY
- interacts with PELP1
- interacts with CBFA2T3
- interacts with SAP30L
Structure:
- belongs to the histone deacetylase family, type 1 subfamily
Compartment: nucleus
Expression:
- widely expressed
- lower levels of expression in brain & lung
Pathology:
- in mice, HDAC2 impairs memory; inhibition of HDAC2 with vorinostat (Zolinza, SAHA) enhances memory [4]
- higher levels of HDAC2 are found in hippocampus from Alzheimer's disease patients
Pharmacology:
- vorinostat (Zolinza, SAHA) inhibits HDAC2
- divalproex inhibits histone deacetylase (HDAC1, HDAC2, HDAC3 ?)
