Function: - NMDA receptor subunit - high Ca+2 permeability - voltage-dependent sensitivity to Mg+2 - mediated by glycine - forms heteromeric channel of: a) zeta subunit (GRIN1) b) epsilon subunit (GRIN2A, GRIN2B, GRIN2C or GRIN2D) c) 3rd subunit (GRIN3A or GRIN3B) - found in a complex with GRIN1 & GRIN3B - found in a complex with GRIN1, GRIN3A & PPP2CB - interacts with PDZ domains of INADL & DLG4 (putative) - interacts with MAGI3 - phosphorylation by PKC enhances activity (putative) Structure: - belongs to the glutamate-gated ion channel (TC 1.A.10) family Compartment: - cell membrane - cell junction, synapse, postsynaptic cell membrane Expression: - mRNA expressed only in forebrain - primarily found in the fronto-parieto-temporal cortex & hippocampus pyramidal cells - lower expression in the basal ganglia
