Function: - may regulate the transcription of specific genes during normal development - may play a role in craniofacial development & digital development, central nervous system development & gastrointestinal tract development - mediates SHH signaling & thus cell proliferation & differentiation - implicated in transduction of sonic hedgehog signalling Structure: - belongs to the GLI C2H2-type Zn+2-finger protein family - contains 5 C2H2-type Zn+2 fingers Compartment: - cytoplasm, nucleus - tethered in the cytoplasm by binding to SUFU - activation & translocation to the nucleus is promoted by interaction with STK36 Expression: - testis, myometrium & fallopian tube Pathology: - defects in GLI1 may be a cause of breast cancer - amplified in glioblastoma cells
