Function: - transcriptional activator - binds to consensus sequence 5'-AGATAG-3' - acts as a transcriptional activator of ANF in cooperation with NKX2-5 (putative) - promotes cardiac myocyte enlargement - methylation at Lys-300 attenuates transcriptional activity - interacts with ZNF260 (putative) - interacts with homeobox domain of NKX2-5 through its C-terminal Zn+2 finger - interaction with JARID2 represses its ability to activate transcription of ANF (putative) - interacts with NFATC4 & LMCD1 - forms a complex with CDK9, CCNT1/cyclin-T1, & EP300 that stimulates hypertrophy in cardiomyocytes Structure: contains 2 GATA-type Zn+2 fingers Compartment: nucleus Pathology: - defects in GATA4 are the cause of a) atrial septal defect type 2 b) atrioventricular septal defect type 4 - defects in GATA4 are a cause of a) ventricular septal defect type 1 b) tetralogy of Fallot
