Fusobacterium nucleatum
Epidemiology:
- present in the human oral cavity
- rarely found in the lower gastrointestinal tract of healthy individual [3]
Pathology:
- may play a role in colorectal carcinoma
- enriched in human colorectal cancer tumors [3]
- stimulates growth of colorectal carcinoma cells
- high tumor Fusobacterium nucleatum loads are associated with recurrence, metastases & poor prognosis [3]
- tumor isolated strains are from the clade Fusobacterium nucleatum animalis C2
- Fusobacterium nucleatum animalis C2 contains a unique adhesion antigen (FadA) on its surface that binds to cell surface E-cadherin, activating beta-catenin signaling & stimulating both inflammation & carcinogenesis
- FadA gene levels in the colon tissue from patients with colorectal adenomas & colorectal adenocarcinomas are > 10-100 times higher than in controls
Management:
- an 11 amino acid inhibitory peptide abolishes FadA-E-cadherin binding & both inflammatory & carcinogenic properties [1]
