Function: - negative regulator of ryanodine receptor Ca2+ release - interacts directly with RYR2 - may contribute to the immunosuppressive & toxic effects of FK506 & rapamycin - rotamases accelerate the folding of proteins - catalyzes the cis-trans isomerization of proline imidic peptide bonds in oligopeptides - inhibited by both FK506 & rapamycin - component of AKAP6/FKBP1B/PKA catalytic/PP1/PP2A/PRKAR2A/RYR2 complex Structure: - belongs to the FKBP-type PPIase family, FKBP1 subfamily Compartment: - cytoplasm . sarcoplasmic reticulum Alternative splicing: named isoforms=2 Expression: - detected in heart muscle (at protein level) - isoform 1 & isoform 2 are ubiquitous with highest levels in brain & thymus Comparative biology: - overexpression of FKBP1b in hippocampus of rats reverses age- associateed memory impairment & neuronal Ca+2 dysregulation [1]
