Function: - seems to plays a role in epithelial tight junction formation - appears early in primordial forms of cell junctions & recruits PARD3 - association of the PARD6-PARD3 complex may prevent interaction of PARD3 with JAM1, thereby preventing tight junction assembly - role in regulating monocyte transmigration involved in integrity of epithelial barrier - role in platelet activation - interacts with the ninth PDZ domain of MPDZ - interacts with the first PDZ domain of PARD3; - association of PARD3 & PARD6B probably disrupts this interaction Structure: - N-glycosylated - belongs to the immunoglobulin superfamily - contains 2 Ig-like V-type domains Compartment: - cell junction, tight junction - cell membrane; single-pass type 1 membrane protein - localized at tight junctions of both epithelial cells & endothelial cells Pathology: - receptor for orthoreovirus - interacts with the orthoreovirus sigma-1 capsid protein (putative)
